Effect of modification of peanut conglutin on IgE and T cell reactivity in adults with peanut allergy
نویسندگان
چکیده
Methods Purified native peanut conglutin, containing Ara h2 and Ara h6, was chemically modified by reduction and alkylation (RA), without or with additional glutaraldehyde treatment (RAGA). Serum of 14 peanut allergic patients (DBPCFC-confirmed) was analyzed for peanut-specific antibodies by IgE immunoblot and IgE/IgG1/IgG4 ELISA. Allergen-induced basophil activation was analyzed by direct basophil activation test (BAT), using whole blood and washed whole blood to remove the plasma. Native conglutin-specific T cell lines were analyzed for proliferation and cytokine production.

Immunoglobulin E cross-reactivity between lupine conglutins and peanut allergens in serum of lupine-allergic individuals.
BACKGROUND Lupine is used increasingly in food products. The development of lupine allergy in peanut-allergic patients is believed to occur as a result of cross-reactivity between lupine and peanut proteins. OBJECTIVE To investigate the degree of immunoglobulin (Ig) E cross-reactivity between allergens in lupine and peanut. METHODS We investigated IgE cross-reactivity between lupine alpha-,...
متن کاملUtility of Specific IgE to Ara h 2 in Italian Allergic and Tolerant Children Sensitized to Peanut
Emerging data suggest that measurement of serum IgE to peanut components can be clinically helpful and more accurate than IgE to whole peanut to predict peanut allergy. Not all studies have used prospective samples, multiple components and oral challenges. Currently, there are no data on this topic involving Italian children. 32 patients (23 males; median age 9 years) with reported history for ...
متن کاملEffect of Heating and Glycation on the Allergenicity of 2S Albumins (Ara h 2/6) from Peanut
BACKGROUND Peanut allergy is one of the most common and severe food allergies, and processing is known to influence the allergenicity of peanut proteins. We aimed to establish the effect of heating and glycation on the IgE-binding properties and biological activity of 2S albumins (Ara h 2/6) from peanut. METHODOLOGY/PRINCIPAL FINDINGS Native Ara h 2/6 was purified from raw peanuts and heated ...
متن کاملHeterogeneous responses and cross reactivity between the major peanut allergens Ara h 1, 2,3 and 6 in a mouse model for peanut allergy
BACKGROUND The relative contribution and the relation between individual peanut allergens in peanut allergic responses is still matter of debate. We determined the individual contribution of peanut proteins to B, T cell and allergic effector responses in a mouse model for peanut allergy. METHODS Mice were immunized and challenged by oral gavage with peanut protein extract or isolated allergen...
متن کاملUse of a basophil activation test as a complementary diagnostic tool in the diagnosis of severe peanut allergy in adults
BACKGROUND Diagnosis of severe peanut allergy is difficult and delays in making an accurate diagnosis may place the patient at risk. Adults with a history of anaphylaxis must strictly avoid any contact with peanuts or products that may contain traces of peanuts. For these persons, conventional evaluations with skin prick testing (SPT) and IgE tests may not be sufficient to assess the risk of an...
